There is no such thing as full inhibition over time.Taken three times a day, orlistat inhibits dietary fat absorption by approximately 30% over a 24 hour period. A single dose will have a shorter, transient effect.PS: There are still plenty of us older Pharmacists with a BSPharm out there working.......
I know the mechanism is inhibiting gastric and pancreatic lipase so less fat can be absorbed but my question is can this be done with a single dose or does it require a period of multiple intakes before the lipase are fully inhibited over time.

I guess what I'm really asking is can a single dose(120mg) of Orlistat inhibit gastric and pancreatic lipease short-term so the effect of a 25-30% reduction of fatty acid absorption in the intestines is achieved or does the compound need to be taken longer for the lipease enzymes to inhibited?

Orilistat (Xenical, Alli) is a reversible inhibitor of lipase meaning it only exerts its effects while the drug is present. There is no such thing as 'full inhibition over time'. Taken three times a day, orlistat inhibits dietary fat absorption by approximately 30% over a 24 hour period. A single dose will have a shorter, transient effect.
