would their spine become straight with the lack of pressure from gravity upon it? This I have wondered for some time now.

No, as their ligaments and muscles have shortened to hold the spine in that place. It's not a gravity thing - people with severe scoliosis do not straighten out when they lie down, for instance.

It is also possible that the G-forces during launch would cause irreparable damage to their spine.

Someone with scoliosis would not be able to lie in the correct position to prevent spinal injury, that's why.
