In a way, yes, but not all of it. Here's the scoop. Niagara Falls is part of the St. Lawrence River drainage which begins around Lake Superior and Lake Michigan. The water from those lakes then flows through Lake Huron, the St. Clair River, Lake St. Clair, and the Detroit River. The Detroit River flows into Lake Erie which is drained by the Niagara River which flows over Niagara Falls. But while the Detroit River is the primary inflow of Lake Erie, it is not the only one. Other major contributors to Lake Erie include the Grand River, the Huron River, the Maumee River, the Sandusky River, the Buffalo River, and the Cuyahoga River. So, as you can see, many rivers other than the Detroit River add to Lake Erie's water before it flows into the Niagara River. So not ALL the water that flows over Niagara Falls went through the Detroit River.
