it says to mash the potato and combine it with water, then filter it through a couple layers of muslin butter (just cloth used to drain water from cheese, if you didn't know) to get the extract. is the extract that contains the enzyme amylase the solid sweet potato part, or is it the liquid that's drained?

Its the liquid part....assuming you sucessfully removed the enzyme from the potato.
I believe this lab also works if you just through the whole potato wedge in there, even before any extraction process.
