When a bacterial cells as taken in a plasmid which as a antibiotic resistant gene to say Amp, why must Amp still remain present when doing an experiment.

If ampicillin is removed then you aren't selecting for the amp resistant strains.... the ampicillin is needed to kill off all the bacterial that are not resistant leaving only the resistant to multiply.
usually an antibiotic resistant bacteria doesn't do as well as a non antibiotic resistant cell so if there's no ampicillin then the non-resistant ones will crowd out the resistant ones and out-compete for resources.
