Can we use PCR to amplify large amounts of DNA to produce intact 100 kb gene region? If not PCR, then what can we use?

you can use bacterial artificial chromosomes(BAC).
The bacterial artificial chromosome's usual insert size is 150-350 kbp, but can be greater than 700 kbp.

yeah I guess. Yeah PCR is never used for cloning. PCR is usually good for amplifying small amounts of DNA. Cloning is you take a diploid nucleus from organism to be clone and insert it into an egg without any nucleus. so you get a exact copy of the genome with the mixing and matching of chromosomes
